Although one cannot directly compare the MICs of different compounds to determine their relative efficacy, the extremely low concentrations of this GC show an effect well below the concentration required of commonly used antibiotics to show a similar effect. These concentrations are also in keeping with concentrations that have been shown to have an antiviral effect of this GC as well.”
Dr. Mazzulli concluded his report by stating, “In the clinical setting, if GC can be shown to be safe and effective, it could provide a breakthrough alternative therapy with potentially significant impact on the practice of family medicine (who initially see most of these common infections) but also in the fields of Otolaryngology, Ophthalmology, and even in the Intensive Care Unit where MDR organisms are a major challenge for treatment.”
Delivery mechanisms like a dry powder inhaler or nasal spray would maximize local concentrations directly at the site of infection and avoid having to administer it systemically (orally or parenterally). Most antimicrobial agents currently used to treat common upper respiratory tract infections such as acute and chronic sinusitis achieve poor concentrations in the sinuses when administered systemically.

The team’s research was part of an EU-funded project, dubbed VANGUARD, which is testing the feasibility of using graphene-oxide scaffolds to help repair and regenerate damaged tissue and organs.
Laura Piddock, director of Antibiotic Action and the public engagement chair at the British Society for Antimicrobial Chemotherapy, told physicsworld.com that the research is “interesting and could provide the basis for a novel preventative agent.
However, there is a very long way to go to show that this can be turned into a medicine that is both safe and efficacious”.

Objective Complications related to infectious diseases have significantly reduced, particularly in the developed countries, due to the availability and use of broad-range antibiotics and wide variety of antimicrobial agents. Excessive use of antibiotics and antimicrobial agents increased significantly the number of multi-drug resistant (MDR) bacteria. This has resulted in a serious threat to public health. The inexorable rise in the incidence of antibiotic resistance in bacterial pathogens, coupled with the low rate of emergence of new clinically useful antibiotics, has refocused attention on finding alternatives to overcome antimicrobial resistance. Novel strategies aiming to reduce the amount of antibiotics, but able to prevent and treat animal and human infections should be investigated, evidenced and approved.
Among the various approaches, the use of graphene and its derivatives is currently considered a highly promising strategy to overcome microbial drug resistance.
In line with this interest in graphene by the European Commission through the graphene ‘flagship’ initiatives, we respond in this consortium by exploring the utility of novel graphene based nanocomposites for the management and better understanding of microbial infections. The anti-microbical potential of the novel graphene based nanomaterials, the possibility of using such structures for the development of non-invase therapies together with the understanding of the mechanism of action will be the main focal points of the proposed project entitled “PANG”, relating to Pathogen and Graphene.

Introduction
The 2030 Agenda for Sustainable Development 1 unites humanity in the pursuit of a common aspiration and a new path of action. The Sustainable Development Goals are for “people, planet and prosperity”. The goals of ending hunger, reducing maternal and infant mortality, and ending epidemics of AIDS, malaria and tuberculosis will require the widespread application of technological breakthroughs in genetics and nanomedicine.
Developing countries may also take advantage of advances in nanotechnology, precision technologies and AI to make plant-derived pharmaceuticals commercially viable for their large populations.
However, the toxicity of these highly volatile compounds remains an issue.
The evaluation of the interactions between pristine and functionalized carbon nanomaterials with animal cells has demonstrated a wide range of possible impacts, from high cell and organism toxicity to no observable effect.419,429
Tangled oxidized multiwalled CNTs were observed to agglomerate into large stable complexes in the intercellular space, undergoing minimal degradation,436 whereas amino-functionalized MWCNTs underwent degradation even at early time points.437 Interestingly, unmodified, ozone-treated, and aryl-sulfonated CNTs did not degrade under these conditions, indicating that acid carboxylation both introduces carboxyl functionality and also damages the tubular graphenic backbone, providing points for subsequent oxidative attacks.438 Carboxyl-functionalized 200 nm graphene sheets also agglomerated in tissues into larger 10 μm structures, gradually developing structural disorder from the edges inward.439 Furthermore, the purity of the nanomaterial may have an effect. Solvents and additives, such as dithio- and diiodooctane,440,441 polydimethylsiloxane,442 or tetrahydrofuran,443 frequently used during synthesis or device assembly may be entrapped in the film or agglomerate, potentially affecting the response of the cell to the nanomaterial.342
Plant peroxidases, e.g. horseradish peroxidase, and animal peroxidases, e.g. myeloperoxidase, eosinophil peroxidase (Figure 33),444,445 and lactoperoxidase, have been shown to catalyze the degradation/biodegradation of carbon nanomaterials.446−448
Strong oxidative activity of horseradish peroxidase (HRP) in the presence of low concentrations of H2O2 is sufficient to initiate biodegradation of carboxylated but not pristine SWCNTs.449 The latter is degraded readily by chemical oxidants (Figure 33), such as hemin, FeCl3, or hypochlorous acid.450 This observation points toward a heterolytic cleavage of H2O2 with HRP as pristine nanotubes remain intact, and a hemolytic cleavage of H2O2 producing free radicals that oxidize pristine SWCNTs.451 It is also likely that the state of agglomeration characteristic of hydrophobic pristine CNTs limits access for the enzymes, thus hindering the effective enzyme−tube interactions.
Prior to the application of these materials, it is important to understand the risks of their migration into food.

Therefore, a lot of research is directed towards the development of test devices that can be used conveniently and reliably at the Point-of-Care (POC) [4]–[7]. It is noteworthy that the World Health Organization (WHO) already identified the place of such methods in diagnostics and have set out the ASSURED (Affordable, Sensitive, Specific, User-friendly, Rapid and robust, Equipment-free and Deliverable to end-users) criteria to be used as a benchmark for developing the most suited test method in resource-constrained situations [8].
Biosensors have the potential to satisfy most/all of these criteria.
They are capable of providing accurate/ precise information on the desired measured health indices as well as sub-clinical level of diseases [9], [10].
6. Conclusion The foregoing shows that significant research progress has been made so far towards diagnostics for POC settings. However, there remains a huge gap in going beyond proof-of-concepts to commercially viable devices i.e. building holistic sensors. Attention must also be given to ensuring accuracy of results and not just sensitivity, specificity, detection limit and other performance metrics.

Graphene and its derivatives are heralded as “miracle” materials with manifold applications in different sectors of society from electronics to energy storage to medicine. The increasing exploitation of graphene-based materials (GBMs) necessitates a comprehensive evaluation of the potential impact of these materials on human health and the environment. Here, we discuss synthesis and characterization of GBMs as well as human and environmental hazard assessment of GBMs using in vitro and in vivo model systems with the aim to understand the properties that underlie the biological effects of these materials; not all GBMs are alike, and it is essential that we disentangle the structure–activity relationships for this class of materials.

The obtained data provide evidence that respirable CNF are quite hazardous, exhibiting similar pulmonary responses to those seen following SWCNT and asbestos exposure. While acute pulmonary inflammation and fibrosis induced by CNF and asbestos were delayed as compared to SWCNT, the systemic immune response elicited by CNF was akin to that observed for asbestos. Exposure to CNF and SWCNT was found to facilitate persistent pulmonary fibrosis along with immune suppression resembling the effects of asbestos, which could potentially promote progression of neoplastic lesions and cancer.

In vivo studies have demonstrated that certain CNTs can cause inflammation, scarring (fibrosis), and granulomas when injected into the lungs of mice [20–23]. In an animal inhalation study, lung discoloration and concentration-dependant lesions (inflammation and granulomas) were observed in the lungs of rodents exposed to relatively low mass doses of multiwall CNTs [20]. Oxidative stress on the pulmonary and cardiovascular systems of rodents exposed to CNTs has been reported as well [24].
When injected into the peritoneal cavity of mice, long CNTs have induced a mesothelioma response [26, 27]. Animal research also indicates that CNTs may be able to translocate within the body and penetrate the pleura in the same way as amphibole asbestos [14, 28]
LITIGATION SCENARIOS Nanotechnology litigation scenarios include consumer and environmental complaints; securities claims; and tort suits. Although not specifically addressed below, insurance disputes are a likely follow-on to any nanotechnology litigation. A. Consumer and Environmental Cases The first wave of nano-litigation could come in the form of consumer or environmental complaints that allege nuisance, breach of implied warranty, fraud, or other theories of liability. In connection with these cases, plaintiffs may seek remediation, injunctive relieve, medical monitoring, and/or monetary damages [38]. A recent decision on bisphenol-A (BPA) consumer class action suggests that such consumer litigation against manufacturers who use nanotechnology in their products could be viable. In the BPA decision, the court allowed the class action to continue despite defendants’ efforts to characterize it as an impermissible “no injury” products liability case. The court reasoned that consumers who learned about BPA’s risks when they had the products and later disposed of them did not obtain the benefit of their bargain [39]. By contrast, where agencies have concluded no health risk exists (e.g., the FDA conclusion that there is no risk from lead used in lipsticks), courts have been more willing to dismiss consumer complaints [39]. Thus, agency decisions could play a central role in any consumer class actions based on undisclosed potential health risks from nanoparticles.
The failure of many companies to engage in product stewardship was the worst mistake of the asbestos experience. We now know better. Responsible corporate behavior has proven critical to consumers and juries alike. Companies – particularly those who use the three nanomaterials discussed herein – are best served by promptly incorporating product stewardship into their business plans.
